Gibberellins (GAs) are the key regulators controlling plant growth, wood production and the stress responses in perennial woody plants. The role of GA in regulating the above-mentioned processes in Eucalyptus remain largely unclear. There is still a lack of systematic identification and functional characterization of GA-related genes in Eucalyptus. In this study, a total of 59,948 expressed genes were identified from the major vegetative tissues of the E. grandis ×E. urophylla using transcriptome sequencing. Then, the key gene families in each step of GA biosynthesis, degradation and signaling were investigated and compared with those of Arabidopsis, rice, and Populus. The expression profile generated using Real-time quantitative PCR showed that most of these genes exhibited diverse expression patterns in different vegetative organs and in response to abiotic stresses. Furthermore, we selectively overexpressed EguGA20ox1, EguGA20ox2 and EguGA2ox1 in both Arabidopsis and Eucalyptus via Agrobacterium tumefaciens or A. rhizogenes-mediated transformation. Though both Arabidopsis EguGA20ox1- and EguGA20ox2-overexpressing (OE) lines exhibited better vegetative growth performance, they were more sensitive to abiotic stress, unlike EguGA2ox1-OE plants, which exhibited enhanced stress resistance. Moreover, overexpression of EguGA20ox in Eucalyptus roots caused significantly accelerated hairy root initiation and elongation and improved root xylem differentiation. Our study provided a comprehensive and systematic study of the genes of the GA metabolism and signaling and identified the role of GA20ox and GA2ox in regulating plant growth, stress tolerance, and xylem development in Eucalyptus;this could benefit molecular breeding for obtaining high-yield and stress-resistant Eucalyptus cultivars.

Nitrogen (N) metabolism plays an important role in plant drought tolerance. 2-(3,4-Dichlorophenoxy) triethylamine (DCPTA) regulates many aspects of plant development; however, the effects of DCPTA on soil drought tolerance are poorly understood, and the possible role of DCPTA on nitrogen metabolism has not yet been explored. In the present study, the effects of DCPTA on N metabolism in maize (Zea mays L.) under soil drought and rewatering conditions during the pre-female inflorescence emergence stage were investigated in 2016 and 2017. The results demonstrated that the foliar application of DCPTA (25?mg/L) significantly alleviated drought-induced decreases in maize yield, shoot and root relative growth rate (RGR), leaf relative water content (RWC), net photosynthetic rate (Pn), stomatal conductance (Gs) and transpiration rate (Tr), and nitrate (NO3?), nitrite (NO2?), soluble protein contents, and nitrate reductase (NR), nitrite reductase (NiR), isocitrate dehydrogenase (ICDH), alanine aminotransferase (AlaAT) and aspartate aminotransferase (AspAT) activities. In addition, the foliar application of DCPTA suppressed the increases of intercellular CO2 concentration (Ci), ammonium (NH4+) and free amino acid contents, and the glutamate dehydrogenase (GDH) and protease activities of the maize. Simultaneously, under drought conditions, the DCPTA application improved the spatial and temporal distribution of roots, increased the root hydraulic conductivity (Lp), flow rate of root-bleeding sap and NO3? delivery rates of the maize. Moreover, the DCPTA application protected the chloroplast structure from drought injury. The data show, exogenous DCPTA mitigates the repressive effects of drought on N metabolism by maintained a stabilized supply of 2-oxoglutarate (2-OG) and reducing equivalents provided by photosynthesis via favorable leaf water status and chloroplast structure, and NO3? uptake and long-distance transportation from the roots to the leaves via the production of excess roots, as a result, DCPTA application enhances drought tolerance during the pre-female inflorescence emergence stage of maize.

Root architecture plays a fundamental role in crop yield, which is sensitive to nitrogen fertilizer. Although it is well studied that nitrogen fertilizer significantly promotes peanut (Arachis hypogaea L.) growth and yield, less information was available on how its root development responds to nitrogen deficiency. In this study, the growth and development of roots were inhibited, as indicated by the significantly decreased root dry weight and length and the lateral root number, especially under 10 days of nitrogen deficiency treatment. The activities and the expression of the genes related to nitrogen assimilation enzymes including nitrate reductase, glutamine synthetase, glutamate dehydrogenase, and glutamine oxoglutarate aminotransferase and the genes encoding the nitrate transporters were significantly decreased under 10 days of nitrogen deficiency treatment, which may lead to a decrease in nitrate content, as indicated by the significantly decreased nitrogen balance index. Transcriptome sequencing revealed a total of 293 (119 up- and 174 downregulated) and 2271 (1165 up- and 1106 downregulated) differentially expressed genes (DEGs) identified after five and ten days of nitrogen deficiency treatments, respectively. Bioinformatic analysis showed that these DEGs were mainly involved in nitrate transportation and assimilation, phytohormone signal transduction, and the lignin biosynthesis pathway. Furthermore, a putative schematic diagram of nitrogen deficiency inhibiting root growth was established, which gives us a better understanding of nitrogen metabolism in peanut roots and a theoretical basis for improving nitrogen use efficiency.

This article proposes a framework P3E for evaluating entity extraction problem over cyber-physical system data. As known by us, this article is the first work focusing on this problem, which is an important step for identifying entities in cyber-physical system data. Directed by the initial entities, utilizing the relaxation and verification techniques, P3E provides a path expression–based solution for entity extraction problem, which has following characteristics. First, using path expressions, P3E provides a condensed presentation for entity locations whose size may get very large when scaling up the data size. Second, requiring only one entity example to indicate the interests, using relaxation technique, P3E can discover other similar entities automatically. Third, by adjusting the example given to P3E, users can specify their own interesting entities and control the entities discovered by P3E. Besides, utilizing the idea of sharing computations, by extending previous automaton techniques, an efficient implementation of P3E is provided. Experimental results are reported, which show that P3E can provide an effective and efficient solution to the entity extraction problem.

Malus baccata Borkh., an apple rootstock, is found to be damaged by oxidation at sub-low root-zone temperature. In previous studies, we have found that exogenous sucrose could alleviate oxidative damage and increase the indole acetic acid (IAA) in roots under sub-low temperature (L). However, the role of IAA in sucrose-induced tolerance to L remains unclear. A pot experiment was conducted to evaluate the effects of exogenous sucrose and IAA synthesis/transport inhibitors (2,3,5-triiodobenzoic acid, TIBA; 4-biphenylboronic acid, BBo) on growth, IAA levels, sugars, and the antioxidant system of M. baccata under L. The results showed that the L treatment decreased IAA contents by 23.69% (48 h) and induced significant increases in root contents of malondialdehyde (MDA) and reactive oxygen (ROS), along with increasing catalase (CAT), ascorbate peroxidase (APX), and glucose-6-phosphate dehydrogenase (G6PDH) activities, while superoxide dismutase (SOD) and monodehydroascorbate reductase (MDHAR) activities first increased (24 h) and then decreased (48 h), and glutathione reductase (GR) and peroxidase (POD) activities significantly decreased. The L treatment also decreased ascorbate/oxidized ascorbate (AsA/DHA), glutathione/oxidized glutathione (GSH/GSSG), and coenzyme II/oxidized coenzyme II (NADPH/NADP+) ratios. Furthermore, the L treatment increased the contents of sucrose, fructose, glucose and sorbitol in the roots and suppressed plant growth. Sucrose pretreatment significantly increased IAA contents (12.42%, 24 h and 14.44%, 48 h) and decreased MDA and ROS contents, which improved the activities of antioxidant enzymes other than APX and increased the contents of AsA, GSH, and NADPH, and increased sucrose, fructose, and sorbitol contents and promoted plant growth. However, the sucrose + TIBA or BBo treatments decreased IAA contents and attenuated or almost abolished the positive effects of exogenous sucrose under sub-low temperature. Our findings indicate that IAA is involved in the sucrose-induced regulation of the antioxidant system in M. baccata roots under sub-low temperature and we provided theoretical references for further study on the adaptability of apple roots to low temperature.

Soil salinity restricts plant growth and productivity. 2-(3,4-dichlorophenoxy) triethylamine (DCPTA) can alleviate salinity stress in plants. However, the mechanism of DCPTA-mediated salinity tolerance has not been fully clarified. We aimed to investigate its role in enhancing photosynthetic capacity, improving water status, maintaining K+/Na+ homeostasis and alleviating salinity stress in maize (Zea mays L.). In present study, maize seedlings were grown in nutrient solutions with a combination of NaCl (0, 150?mM) and DCPTA (0, 20, 100, and 400?μM). And photosynthesis, water status, ion homeostasis and the expression of genes involved in ion uptake and transport were evaluated in the maize seedlings. The results demonstrated that DCPTA alleviated the growth inhibition of maize seedlings exposed to salinity stress by increasing the net photosynthetic rate (Pn) and the quantum efficiency of photosystem II (PSII) photochemistry. DCPTA improved the root hydraulic conductivity, which help maintained the water status. A relatively high K+ concentration but a relatively low Na+ concentration and the Na+/K+ ratio were observed in the presence of DCPTA under salinity stress. Additionally, DCPTA altered the expression of four genes (ZmSOS1, ZmHKT1, ZmNHX1 and ZmSKOR) that encode membrane transport proteins responsible for K+/Na+ homeostasis. DCPTA improved the salinity tolerance of maize may be associated with enhanced photosynthetic capacity, maintenance of water status and altered expression of genes involved in ion uptake and transport.

Numerous experimental simulations with different warming scenarios have been conducted to predict how algae will respond to warming, but their conclusions are sometimes contradictory to each other. This might be due to a failure to consider interspecific interactions. In this study, the dominant diatom species in a seasonal succession were isolated and verified to adapt to different temperature ranges by constant temperature experiment. Both unialgal and mixed cultures were exposed to two fluctuant temperature treatments that simulated the temperature variations from early spring to summer, with one treatment 4 °C higher (warming scenario) than the other. We found that the specific response of diatoms to warming was affected by interspecific interactions. Spring warming had no significant effect on eurythermal species and had a positive effect on the abundance of warm-adapted diatom species, but interspecific interactions reduced this promotional effect. Cold-adapted species had a negative response to spring warming in the presence of other diatom species but had a positive response to early spring warming in the absence of interspecific interactions. In addition, warming resulted in the growth of all diatom species peaking earlier in unialgal cultures, but this effect could be weakened or amplified by interspecies interactions in mixed cultures. Our results suggest that the specific diatom species with different optimal growth temperature ranges responding to warming were expected if there were no interspecific interactions. However, in natural environments, the inevitable and complex interspecific interactions will influence the responses of diatoms to warming. This important factor should not be ignored in the prediction of organism responses to climate warming.

Heat shock protein 90 (HSP90) functions as a well-known onco-protein to regulate protein conformation, stability and degradation. Pyruvate kinase M2 (PKM2), a critical regulator of the metabolism, growth and metastasis of cancer cells, has been confirmed to be overexpressed in various human cancer including hepatocellular carcinoma (HCC). However, the molecular mechanisms underlying the oncogenic functions of HSP90 and PKM2 overexpression in HCC remain unknown. The expression of HSP90 and PKM2 in HCC specimens and cells were detected by immunoblotting and immunostaining. The interaction between HSP90 and PKM2 was confirmed by tandem affinity purification, co-immunoprecipitation and Glutathione S transferase (GST)-pulldown assay. In this study, we found that HSP90 could bind to PKM2 and subsequently increased PKM2 abundance in HCC cells. Immunohistochemistry (IHC) staining showed that HSP90 level was positively correlated with PKM2 level in HCC tissues. Mechanistically, HSP90 was found to increase the phosphorylation of PKM2 at Thr-328. Protein kinase glycogen synthase kinase-3β (GSK-3β) formed a protein complex with HSP90 and PKM2, and directly mediated Thr-328 phosphorylation of PKM2 induced by HSP90. Thr-328 phosphorylation was critical for maintaining PKM2 stability and its biological functions in regulating glycolysis, mitochondria respiration, proliferation and apoptosis. Functionally, we found that HSP90 promoted the glycolysis and proliferation and inhibited apoptosis of HCC cells in a PKM2 dependent manner. In vivo experiments disclosed that PKM2 was required for the promoting effects of HSP90 on the growth of HCC cells in mice. Furthermore, we demonstrated that positive expression of HSP90 and PKM2 was correlated with poor clinicopathological features including high alpha fetoprotein (AFP) level, large tumor size, portal vein tumor thrombus (PVTT) and advanced tumor-node-metastasis (TNM) stage. Furthermore, we demonstrated that positive expression of HSP90 and PKM2, and a combination of these proteins could strongly predict the poor prognosis of HCC patients. We suggest that HSP90 potentiates the glycolysis and proliferation, reduces the apoptosis and thus enhances the growth of HCC cells through PKM2.
